چکیده

Transcripts from the Xenopus gene GS17 are absent from oocytes and first appear at the midblastula transition (MBT), about 8 hr after fertilization. Injection experiments with deletions of the GS17 gene and GS17-beta-globin hybrid genes show that a relatively short sequence of 74 base pairs is sufficient to activate, in cis, transcription at the MBT. This 74-base sequence is located about 700 bases upstream of the GS17 promoter and has the properties of an enhancer element.

Histone titration against the genome sets the DNA-to-cytoplasm threshold for the Xenopus midblastula transition.

During early development, animal embryos depend on maternally deposited RNA until zygotic genes become transcriptionally active. Before this maternal-to-zygotic transition, many species execute rapid and synchronous cell divisions without growth phases or cell cycle checkpoints. The coordinated onset of transcription, cell cycle lengthening, and cell cycle checkpoints comprise the midblastula t...

DNA sequence-specific binding activity of the heat-shock transcription factor is heat-inducible before the midblastula transition of early Xenopus development.

We have examined the activity of the Xenopus heat-shock transcription factor (HSF) in extracts from stressed and unstressed embryos at various stages of development using DNA mobility shift analysis. A specific interaction between HSF and a synthetic oligonucleotide corresponding to the proximal heat-shock element (HSE) of the Xenopus HSP70B gene was greatly enhanced in heat-shocked embryos com...

Brg1 chromatin remodeling ATPase balances germ layer patterning by amplifying the transcriptional burst at midblastula transition

Zygotic gene expression programs control cell differentiation in vertebrate development. In Xenopus, these programs are initiated by local induction of regulatory genes through maternal signaling activities in the wake of zygotic genome activation (ZGA) at the midblastula transition (MBT). These programs lay down the vertebrate body plan through gastrulation and neurulation, and are accompanied...

The pattern of early cleavage of the marsupial frog Gastrotheca riobambae.

Comparison of early development of the marsupial frog Gastrotheca riobambae with Xenopus laevis suggests that the cleavage pattern of Xenopus and the tight coupling of events at the midblastula transition are features of the accelerated development of small amphibian eggs with aquatic reproduction, rather than generalized features of amphibian development. The large eggs of the marsupial frog G...

Nuclear Size Scaling during Xenopus Early Development Contributes to Midblastula Transition Timing

Early Xenopus laevis embryogenesis is a robust system for investigating mechanisms of developmental timing. After a series of rapid cell divisions with concomitant reductions in cell size, the first major developmental transition is the midblastula transition (MBT), when zygotic transcription begins and cell cycles elongate. Whereas the maintenance of a constant nuclear-to-cytoplasmic (N/C) vol...
